I will discuss the synthesis of modified macrocyclic peptides using enzymes. The patellamide pathway is an exemplar for ribosomal peptide biosynthesis. We have characterised the enzymes in this pathway and shown synthetic utility. We have used these enzymes in synthesis, engineering some to overcome their limitations. We have recently characterised two new macrocyclases that represent a promising set of tools. I will discuss an unusual enzymatic reaction that has value in peptide modification.
